惩处干部违法违纪与纠正不正之风是中纪委上年度工作的两大关键词。第十八届中央纪委第二次全会提出：“既坚决查处领导干部违纪违法案件，又切实解决发生在群众身边的不正之风和腐败问题。”
在查处领导干部方面，2012年12月初，四川省委原副书记李春城因涉嫌严重违纪接受组织调查，成为十八大后首个“落马”的省部级干部。
此后，广东省委原常委、统战部原部长周镇宏，国家发改委原副主任、国家能源局原局长刘铁男，安徽省政府原副省长倪发科，内蒙古自治区原党委常委王素毅，广西政协原副主席李达球，中国农业银行股份有限公司原执行董事副行长杨琨等一批省部级领导干部受到查处。
各地区各部门也查处了一批在本地区本部门有影响的大案要案。比如，安徽省纪委查办了省国土资源厅原厅长陈良纲严重违纪违法案件，广东省纪委查办了深圳市原副市长梁道行严重违纪违法案件。

针对群众身边的腐败问题，全国纪检监察机构也查办了一批农村案件和重点部门案件。
2013年，全国纪检监察机关查办发生在农村的案件8.3万件，处分8.7万人；查办了一批在人力资源和社会保障、医疗卫生、环境保护、国土资源、教育等部门的案件，处分1.5万人，比上年同期增长27.0%。同时，查处了一批职级不高、涉案金额巨大、群众反应强烈的“小官巨贪”案件。
为了加大查办案件力度，2013年初，中央纪委、监察部增设了2个纪检监察室。参照这一做法，各地纪检监察机关相应做了调整。截至2013年10月底，已有17个省(区、市)纪委新设立1~2个纪检监察室。
